Confocal Wire Myograph

Confocal Wire Myograph

Overview:

The DMT120CW Confocal Wire Myograph is specifically designed for obtaining highly resolved images of fluorescent dyes within living tissues (> 60 µm) using laser-scanning confocal microscopy (LSCM). In combination with LSCM, simultaneous measurements of isometric forces and intracellular Calcium or pH can be made. The DMT120CW includes a control unit with temperature control, built-in high-resolution force transducer and micrometer.

The control unit has analog output which can be connected directly to a PowerLab BNC input. The Confocal Wire Myograph System is also available and includes a PowerLab data acquisition system, LabChart Pro and DMT120CW. Optical components are NOT included.

When purchasing the DMT120CW the following items are included:

Myo- Interface Unit

  • Myograph connection cable with a temperature probe
  • Power cord (The shape of the AC plug varies by country; be sure that the plug has the right shape for your location)

Confocal Wire Myograph Unit

  • Calibration kit (including bridge , balance and 2 gram weight)
  • Chamber cover
  • Chamber volume 10 ml

Accessories

  • 1 roll of 40 μm stainless steel wire
  • 1 tube of high vacuum grease
  • 1 tube of grease for linear slides
  • 4 spare screws for mounting of jaws
  • 10 extra myograph window glasses
  • 2 Allen keys
  • 1 small screwdriver
  • 40 mm funnel

Manuals

  • User manual for the Confocal Wire Myograph System Model 120CW
  • Procedures for investigation of small vessels using small vessel myograph, by Professor M. J. Mulvany, Department of Pharmacology, Aarhus University,Denmark

Warranty Exclusion

  • DMT force and pressure transducers, either separately or part of myograph systems manufactured by DMT A/S, are disclaimed from any warranty

Features:

  • Single channel wire myograph (10ml)
  • Bath design allows easy access for high numerical aperture lenses used in standard upright confocal microscopy
  • Reverse mounting supports allow vertical positioning of the isolated blood vessel

Applications:

The DMT120CW Confocal Wire Myograph is specifically designed for obtaining highly resolved images of fluorescent dyes within living tissues using laser-scanning confocal microscopy. It is ideal for physiological and pharmacological investigations using arterial microvessels, and a variety of other tissue types such as small veins, renal arcuate arteries, bronchi and ureter. The technique, is suited to vessels with internal diameters > 60 μm.

Parameters which can be measured include:
  • Normalized internal diameter
  • Measuring tissue wall thickness
  • Isometric (force) responses to drugs (agonists/antagonists)
  • Simultaneous measurements of isometric response and cell membrane potential
  • Simultaneous measurements of isometric responses and ion concentrations inside the cell
  • Analysis of tissue fixed under clearly defined conditions for microscope measurements
  • Fluorescent Microscopy; Intracellular [Ca2+] or pH.
For more information see the Wire Myography application page.
