Radnoti Specialized Tissue Baths
Overview
A range of high-quality Radnoti Tissue Organ Baths are available, which are specially designed to accommodate specific tissue and study requirements.
Radnoti 1650 Series “Q” Quiet Tissue Organ Baths
Oxygenation of tissues, especially cardiac muscle is a requisite in maintaining normal physiological and biochemical responses. However, aeration can introduce mechanical noise due to impinging of gas bubbles on the suspended tissue. This mechanical noise can obscure the force measurements collected from small tissue samples, such as papillary muscles. Hence, the Radnoti 1650 Series “Q” Quiet Tissue Organ Baths were designed to solve this issue, which virtually eliminates gas bubble interference.
Its design incorporates many desirable features of the Radnoti 1660 Series Hi-Tech Tissue Organ Baths. Utilizing the basic principles from a design by the eminent physiologist J.R. Blinks (J. Appl. Physiol. [1965] 20:755-7), the bath is a dual-chambered U-tube with a bridge connection midway between the two chambers. One chamber is used for gassing and has a removable Oxygenating Tube with Teflon Needle Valve to permit fine adjustments to the gas flow. A circulating flow in the solution is generated by the rising gas bubbles during gassing. The tissue sample is placed in the other chamber. This design efficiently separates gas bubbles from the tissue chamber, yet it allows the oxygen-enriched solution to circulate through both chambers.
Therefore, it is also ideal in cases where the researcher must resolve extremely small tissue force measurements or where direct aeration would cause problems. Other potential uses would be side-by-side comparisons of tissues in the same bath, such as vessel rings with and without endothelium, etc.
Other features include:
- A water jacket outer compartment that uses Radnoti Quick Disconnect fittings to ensure leak-resistant water jacket connections, while providing easy attachment or detachment
- A Luer overflow port that prevents overflow in protocols that require the constant addition of solution via a pump or reservoir, and permits solution sampling for analysis
- Has a bottom Luer drain port that can be fitted with a 1-Way Tap to permit rapid flushing
- Allows complete removal of bath solution, which permits both effective tissue washing and complete collection of endogenously released substances
- Works either a corresponding sized Radnoti Glass Tissue Hook or a Radnoti Tissue Ring Support.
- Accommodates stimulating electrodes that may be inserted into either or both chambers.
- Available in 25 or 50 mL bath volumes
They attach to Radnoti Lab Stands via Single Ring Clamps and Universal Stand Clamps.
Radnoti 1583 Series Low Profile Non Aerating Tissue Bath
A 50 mL Radnoti tissue organ bath that is designed to maintain tissue samples or solutions at a desired non-ambient temperatures, but do not require gassing or aeration. It attaches to a Radnoti Lab Stand via a Single Ring Clamp and Universal Stand Clamp. Similarly, it has a water jacket outer compartment that uses Radnoti Quick Disconnect fittings, a Luer overflow port, a bottom Luer drain, and is compatible with either a corresponding sized Radnoti Glass Tissue Hook or a Radnoti Tissue Ring Support.
Radnoti 1660 Series Tissue Organ Baths for Large Organ or Single Cell Isolation
Specialized Radnoti 1660 Series Hi-Tech Tissue Organ Baths that are designed to either accommodate larger organs, such as adrenal glands, hearts, livers and kidneys, or for performing cell isolation from such organs. They are designed with a water jacket outer compartment that uses Radnoti Quick Disconnect fittings, an in-built pre-warming coil within the water jacket section of the tissue chamber, a Luer overflow port, a bottom side Luer drain port, and a removable Oxygenating Tube with Teflon Needle Valve.
They are available in two bath volumes, 200 mL (45 mm Depth) and 550 mL (100 mm Depth), both having a 125 mm outer diameter, and a 95 mm inner diameter. However, each has slightly different designs. The pre-warming coil of the 200 mL delivers solutions to a Luer entry port located two thirds of the way up the bath interior. The Luer fitting permits attachment of a cannula which is then placed into the organ. The 550 mL bath has a pre-warming coil with bubble trap and a Luer lock fitting.
Radnoti 1584 Series Preparatory Tissue Baths
In isolation protocols, often the removal of the tissue from its donor to the insertion of the tissue into an organ bath results in the tissue being exposed to temperature and oxygen variations, and dissection trauma, which can lead to loss of tissue viability and compromised physiological and pharmacological properties. The Radnoti Preparatory Tissue Baths are designed to minimize variability and tissue damage during that critical period. They feature a water jacket outer compartment that uses Radnoti Quick Disconnect fittings, and an in-built side-mounted gas dispersion tube that provides gassing to maintain tissue viability.
Additionally, the shallower version of the bath provides a stable, convenient preparatory surface that can even be positioned beneath a dissecting microscope. If required, the tissue may be pinned to the bottom of the bath using a 4-6 mm paraffin or Sylgard coating (ensure the opening of the gas dispersion tube is temporarily sealed by a tape before application and unseal after the coating hardens). If necessary, the bath may be chilled by the circulation of cold water or an antifreeze solution or placed upon crushed ice.
Note: All glassware are fabricated out of chemically inert and durable borosilicate glass, which is fully autoclavable The actual volumes of all baths are greater than stated volumes to allow for displacement of supports, accessories and samples. Additionally, if ordering the bath individually, components, such as Radnoti Quick Disconnect tubing and fittings will be required and are purchased separately.
